I believe it's considered a rare spawn. I know I did three runs on the Ectoplasm quest before a ghost dropped on but found the prismatic slime fairly quickly.

In my case, I spent all day in the mines looking for the guy, and didn't appear until the 5th of the 7 days you have. It's a rare spawn, keep insisting.

Bumping this bug. I searched the mountain mine for days while doing this quest and found no prismatic slime. On the final day of the quest, I used a mod to freeze time in caves. I spent hours looking on almost every floor, resetting and searching again.......no prismatic slime at all. I just finally had to give up and fail the quest. Was I in the wrong mine? (it really cant be that rare!)

I'm also pretty disappointed about this quest. I spent my whole 7 days to search for that slim, searching carefully each floor one by one (pretty sure I searched every 120 floors) but I didn't find anything...
I understand that those quest are designed to be difficult, but this one is maybe little bit too unfair :(

I've also attempted this quest twice now and--while I may not have been as thorough as some of you--I have also been unable to locate the Prismatic Slime of legend. Honestly, I just hit any slime I see hoping I just didn't catch the color change, but still no luck.

I tried at least another 100 floors by using the wiki advice (go to floor 5, check, exit, return to floor 5 etc.), still nothing.
If it can help, I'm using a custom random seed filled by 0's, and I'm in fall 8-14 (14 for my loop test)

I don't think it is a bug (but impossible to be sure actually). But I think this quest is too difficult because the probability to encounter the slime is too small.
Maybe it is done on purpose, but it is frustrating :/

After another run of ~150 tries (for a total of more than 350), I start to think it could be a bug for real...
This has been addressed with the 1.5.3 update, please mention if you're seeing a change! From the patch notes:

After a few tries, I'm proud to tell you I finally manage to have that slime ! 
It took me only about 3 hours in game (so, about 10 floors to explore) with "little bit annoyed" spirits

I *finally* killed a prismatic slime on the 83rd floor of The Mines and it dropped a Yeti Tooth instead of the jelly.

Are you sure it was prismatic ? Did you update to 1.5.3 ?
According to the changelog, this bug should not happen anymore
Just to confirm, if you click the ? on the title screen, what version does it list in the bottom left? And are you playing with any mods?

No mods and it says 1.5.
Sounds like you need to update you game, you will want to launch it through Steam or GOG (whichever one you bought the game through) in order to update to 1.5.4
