Would it work? Sure. Would it become a popular competitive category? Probably not, but you never know. If you think it sounds like fun, go for it. That's really all you need. 
People can - and do - speedrun anything, even finding creative ways to use otherwise not-particularly-speedrun-friendly games, like the 'Homepour' run. For those not in the know, this is trying to complete a speedrun of Silent Hill Homecoming whilst only playing it during the cutscenes of Silent Hill Downpour. (Downpour is not popular for speedrunning due to having an hour and a half of unskippable cutscenes. Which is about the same time as a good speedrun of Homecoming, so you can see where the logic came from.

Not a speedrunner myself, I've done the odd strange challenge run but I mostly just watch them and their technical explanations on youtube.

They also do often have unofficial joke categories (some of which have been adapted to official-joke categories by being part of a joke tab on the speedrun.com leaderboard), though they’re usually short runs like urchin hat%.
